In 1982 Sergei Aleksashkin graduated from the Saratov Conservatory and later studied in Italy for two years at the Teatro della Scala. In 1989 he joined the Kirov Opera, with which he regularly tours.

He first performed in Britain in 1991 at the Edinburgh Festival and in the same year sang with Placido Domingo in Otello in Spain. He performed at the Metropolitan Opera, New York in 1992 and in 1993 made his Japanese debut in Boris Godunov, broadcast by NHK, followed by performances at the Vienna Konzerthaus of Shostakovich's Symphony No.14, recorded by Denon, Rachmaninov's Miserly Knight in Dresden and a European tour of The Damnation of Faust with Gergiev. In 1994 Sergei Aleksashkin sang in Boris Godunov in the Salzburg Festival with Abbado, returned to Japan for Shostakovich's Symphony No.13 and in 1995 took this work on a long-term international tour of major festivals, conducted by Solti and recorded by Decca. In 1997 he sang the role of Don Basilio (The Barber of Seville) for the Royal Opera House at the Shaftesbury Theatre, London.

He toured with the Kirov-Maryinsky Company in Germany, Italy, Spain, Japan, UK (Edinburgh Festival 1991) and the USA (the Met 1992). In 1998-99 he made his UK debut with the BBC Philharmonic performing Mussorgsky’s Songs and Dances of Death conducted by Solti. Other engagements in the West included Boris Godunov in Frankfurt The Miserly Knight (Dresden); Prince Igor (Vienna); Tchaikovsky's Enchantress (Concertgebouw); La Damnation de Faust (Rome with the Academy and Chorus of Santa Cecilia); Basilio in Il barbiere di Siviglia (Royal Opera House, Covent Garden); Pimen in Boris Godunov (Washington, Lyon); Dosifei in Khovanshchina, Kutuzov in War and Peace, Kotchubei in Mazeppa under Gergiev (Kirov at the ROH); The General in The Gambler (Metropolitan Opera), Kutuzov at the Teatro Real, Madrid and King René in Iolanta (Netherlands Opera, Châtelet).

His opera repertoire also includes: Mozart (Don Giovanni, Leporello, Sarastro); Verdi (Filippo, Inquisitore, Procida (I Vespri Siciliani), Fiesco, De Silva, (Ernani), Ramphis, Padre Guardiano); Rossini (Don Magnifico); Gounod (Mephistopheles); Wagner (The King, Lohengrin); Glinka (Ivan Susanin, A Life for the Tsar, Ruslan, Farlaf, Ruslan and Ludmilla); Mussorgsky (Cherevik's Crony, Sorochintsky Fair); Rachmaninov (Aleko); Rimsky Korsakov (Ivan the Terrible, The Maid of Pskov, Prince Yuri, The Invisible City of Kitezh, Salieri, Mozart and Salieri, Maliuta, Sobakin, The Tsar's Bride, Tsar of the Sea, Sadko); Offenbach (Lindorf, Miracle, Coppelius, The Tales of Hoffmann) and Shostakovich (Boris Timofeevich, Katerina Ismailova).

Aleksashkin's concert repertoire includes Beethoven (Symphony No. 9); J.S. Bach (Magnificat, B Minor Mass); Berlioz (Mephistopheles, The Damnation of Faust; Friar Laurence, Romeo et Juliet); Dvorak (Biblical Songs); Mahler (Symphony No. 8); Mozart (Requiem); Verdi (Requiem); Shostakovich's Symphonies No. 13 and 14 and his Sonet by Michaelangelo, plus many songs and romances by Russian and western European composers.

He has made numerous recordings with conductors such as Gergiev, Solti, Rozhdestvensky, Temirkanov and Järvi and with labels such as Deutsche Grammophon, Philips, Decca and BMG.

In the summer of 2006 he sang Mendoza in Betrothal in a Monastery in his debut at the Glyndebourne Festival and King Dodon in The golden cockerel at the Mikkeli Festival in Finland. Most recently he sang Prince Gremin in Eugene Onegin at the Metropolitan Opera, New York, and the Dvorak Requiem in Lisbon.
